The Mushroom Rock Road is a pathway that you can take from the Djose Highroad / Mi’ihen Highroad area.

Traveling through the valley and participating in the first battle against Sin with the Al Bhed and their Machina.
Late in the game, after you have obtained the airship, the Dark Magus Sisters will ambush travelers near the Mi’ihen Highroad end of the road. You can avoid the encounter by entering the area from the Djose Highroad side.
