The first trip to the Lhusu Mines occurs after the team reaches Bhujerba for the first time. Most of the zone, including all areas beyond Transitway 2 (and areas of Sites 2 and 3), is inaccessible. There are additional visits back to the Lhusu Mines later on in the game to complete several Hunts.
